SYK (Spleen-Associated Tyrosine Kinase) is a non-receptor tyrosine kinase belonging to the Syk family, closely related to ZAP-70. SYK is activated downstream of immunoreceptor tyrosine-based activation motifs (ITAMs) following engagement of B-cell receptors (BCR), Fc receptors, and integrins. Upon ITAM phosphorylation by Src-family kinases, the tandem SH2 domains of SYK engage dually phosphorylated ITAMs, triggering SYK autophosphorylation at Y352 and Y525/Y526, driving full catalytic activation. Key substrates include BLNK, PLC-gamma2, PI3K regulatory subunits, and VAV proteins, propagating NF-kB, MAPK, and calcium signaling cascades. SYK is central to B-cell development, mast cell degranulation, platelet activation, and innate immune signaling. Dysregulation drives B-cell malignancies including diffuse large B-cell lymphoma (DLBCL), chronic lymphocytic leukemia (CLL), mantle cell lymphoma, and follicular lymphoma. Elevated SYK activity also contributes to autoimmune diseases such as rheumatoid arthritis and immune thrombocytopenic purpura, making it a high-priority therapeutic target in oncology and immunology.
SYK assays present several challenges: high physiological ATP concentrations in immune cells require assays tolerant of millimolar ATP, yet many luminescence-based platforms such as ADP-Glo suffer signal compression at high [ATP], underestimating potency of ATP-competitive inhibitors. HTRF and LANCE antibody-based assays introduce variability from epitope competition and require fixed endpoints that miss mechanistic detail. Radiometric formats are operationally burdensome and generate radioactive waste. For covalent SYK inhibitors, traditional endpoint formats cannot capture the full kinact/KI profile. PhosphoSens CSox-based peptide substrates enable continuous, real-time fluorescent monitoring of SYK-mediated phosphorylation at physiological ATP, generating complete progress curves. This allows precise Km(ATP) determination, accurate IC50 measurement without endpoint artifacts, and direct kinact/KI characterization of irreversible inhibitors — all in a homogeneous, antibody-free, wash-free format compatible with high-throughput screening.
