PhosphoSens SYK kinase assay kit — continuous real-time fluorescence

About SYK

SYK (Spleen-Associated Tyrosine Kinase) is a non-receptor tyrosine kinase belonging to the Syk family, closely related to ZAP-70. SYK is activated downstream of immunoreceptor tyrosine-based activation motifs (ITAMs) following engagement of B-cell receptors (BCR), Fc receptors, and integrins. Upon ITAM phosphorylation by Src-family kinases, the tandem SH2 domains of SYK engage dually phosphorylated ITAMs, triggering SYK autophosphorylation at Y352 and Y525/Y526, driving full catalytic activation. Key substrates include BLNK, PLC-gamma2, PI3K regulatory subunits, and VAV proteins, propagating NF-kB, MAPK, and calcium signaling cascades. SYK is central to B-cell development, mast cell degranulation, platelet activation, and innate immune signaling. Dysregulation drives B-cell malignancies including diffuse large B-cell lymphoma (DLBCL), chronic lymphocytic leukemia (CLL), mantle cell lymphoma, and follicular lymphoma. Elevated SYK activity also contributes to autoimmune diseases such as rheumatoid arthritis and immune thrombocytopenic purpura, making it a high-priority therapeutic target in oncology and immunology.

Why PhosphoSens for SYK?

SYK assays present several challenges: high physiological ATP concentrations in immune cells require assays tolerant of millimolar ATP, yet many luminescence-based platforms such as ADP-Glo suffer signal compression at high [ATP], underestimating potency of ATP-competitive inhibitors. HTRF and LANCE antibody-based assays introduce variability from epitope competition and require fixed endpoints that miss mechanistic detail. Radiometric formats are operationally burdensome and generate radioactive waste. For covalent SYK inhibitors, traditional endpoint formats cannot capture the full kinact/KI profile. PhosphoSens CSox-based peptide substrates enable continuous, real-time fluorescent monitoring of SYK-mediated phosphorylation at physiological ATP, generating complete progress curves. This allows precise Km(ATP) determination, accurate IC50 measurement without endpoint artifacts, and direct kinact/KI characterization of irreversible inhibitors — all in a homogeneous, antibody-free, wash-free format compatible with high-throughput screening.

Frequently Asked Questions: SYK Assays

Can PhosphoSens SYK assays be run at physiological ATP concentrations?

Yes. PhosphoSens CSox-based assays are fully compatible with physiological ATP concentrations (1-2 mM), unlike ADP-Glo or luminescence platforms that saturate or compress signal at high [ATP]. This enables accurate determination of Km(ATP) and physiologically relevant IC50 values for ATP-competitive SYK inhibitors.

How does the PhosphoSens assay support characterization of covalent SYK inhibitors?

The continuous, real-time progress curve format of PhosphoSens allows direct measurement of time-dependent inhibition kinetics required to extract kinact and KI for covalent or irreversible SYK inhibitors. Endpoint assays such as HTRF or ADP-Glo cannot capture this time-resolved inhibition profile, leading to inaccurate potency estimates for covalent compounds.

What substrate does AssayQuant use for SYK kinase activity measurements?

AssayQuant offers optimized CSox-based peptide substrates derived from validated SYK substrate sequences, designed to be phosphorylated selectively by SYK with high sensitivity. The fluorescent signal increases continuously upon phosphorylation, enabling real-time kinetic readout without antibodies, radioactivity, or secondary detection reagents.
