Cyclin-dependent kinase 1 (CDK1) is the founding and most essential member of the CDK family, functioning as the master regulator of mitotic entry and progression in eukaryotic cells. CDK1 is catalytically inactive as a monomer and requires association with cyclin partners — primarily Cyclin B1/B2 for M-phase entry and Cyclin E1 for S-phase regulation — as well as activating phosphorylation by CDK-activating kinase (CAK) at Thr161. CDK1/CyclinE1 complexes contribute to G1/S transition control, licensing origin firing and coordinating with CDK2 functions. Key substrates include Rb family members, Lamins, condensin, separase inhibitory proteins, and numerous centrosomal and spindle assembly factors. CDK1 sits at the convergence of the RB/E2F, DNA damage checkpoint, and Wnt signaling pathways. Dysregulation of CDK1 activity — through cyclin overexpression, loss of CIP/KIP inhibitors, or checkpoint abrogation — is pervasive across solid tumors and hematologic malignancies, making CDK1 a compelling oncology drug target whose inhibition forces mitotic catastrophe in cancer cells.
CDK1/CyclinE1 drug discovery presents distinct challenges: the kinase shares high structural homology with CDK2 requiring exquisite selectivity profiling, and its activity is tightly regulated by phosphorylation state and cyclin stoichiometry, making endpoint assays prone to artifacts from incomplete reaction capture. ADP-Glo and HTRF endpoint formats miss burst-phase kinetics and covalent engagement, while radiometric methods lack throughput and real-time resolution.
