About AURKA

Aurora Kinase A (AURKA) is a serine/threonine kinase belonging to the Aurora family, which also includes AURKB and AURKC. AURKA localizes to centrosomes and spindle poles, where it orchestrates mitotic entry, centrosome maturation, bipolar spindle assembly, and cytokinesis. Activation requires autophosphorylation at Thr288 within the activation loop, potentiated by binding partners including TPX2, which shields Thr288 from PP1-mediated dephosphorylation. Key substrates include LATS2, PLK1, BRCA1, CDC25B, and Eg5, linking AURKA to G2/M checkpoint regulation and DNA damage response. AURKA is amplified or overexpressed in breast, colorectal, ovarian, pancreatic, and hematologic malignancies, driving chromosomal instability and resistance to taxane-based therapy.
