About CDK6/CycD1

Cyclin-dependent kinase 6 (CDK6) is a serine/threonine kinase belonging to the CDK family that requires association with a D-type cyclin, most notably Cyclin D1, for catalytic activation. The CDK6/CyclinD1 complex is a master regulator of the G1-to-S phase transition of the cell cycle. Upon mitogenic stimulation, Cyclin D1 accumulates and binds CDK6, enabling phosphorylation of the retinoblastoma protein (Rb) and related pocket proteins p107 and p130. Rb hyperphosphorylation releases E2F transcription factors, driving expression of S-phase entry genes. Beyond cell cycle control, CDK6 exerts transcriptional functions independent of Rb, regulating NF-kB, RUNX1, and VEGF-A.
