About BRSK2

BRSK2 (Brain-specific serine/threonine kinase 2), also known as SAD-A, belongs to the CAMK-like (CAMKL) superfamily and is closely related to BRSK1, MARK, and AMPK family kinases. Activated by LKB1-mediated phosphorylation on its T-loop threonine (Thr174), BRSK2 is a master regulator of neuronal polarity, establishing axon-dendrite identity during neurodevelopment. Key substrates include tau, MAP2, and components of the WNT signaling pathway including Dvl proteins. Beyond the CNS, BRSK2 plays roles in pancreatic beta-cell function, insulin secretion, and cell cycle regulation via CDC25 phosphorylation. Dysregulation of BRSK2 has been linked to neurodevelopmental disorders, autism spectrum disorder, schizophrenia, and metabolic diseases.
