BRSK2 (Brain-specific serine/threonine kinase 2), also known as SAD-A, belongs to the CAMK-like (CAMKL) superfamily and is closely related to BRSK1, MARK, and AMPK family kinases. Activated by LKB1-mediated phosphorylation on its T-loop threonine (Thr174), BRSK2 is a master regulator of neuronal polarity, establishing axon-dendrite identity during neurodevelopment. Key substrates include tau, MAP2, and components of the WNT signaling pathway including Dvl proteins. Beyond the CNS, BRSK2 plays roles in pancreatic beta-cell function, insulin secretion, and cell cycle regulation via CDC25 phosphorylation. Dysregulation of BRSK2 has been linked to neurodevelopmental disorders, autism spectrum disorder, schizophrenia, and metabolic diseases. Somatic mutations and altered expression are observed in colorectal, breast, and pancreatic cancers, implicating BRSK2 as an oncology target. Its position downstream of the tumor suppressor LKB1 further elevates its therapeutic relevance in precision oncology and neuroscience drug discovery programs.
BRSK2 presents several assay challenges: low intrinsic activity in recombinant preparations, requirement for LKB1-dependent activation, and sensitivity to ATP concentration that complicates conventional endpoint assays. ADP-Glo and HTRF methods require fixed endpoint reads, missing the early progress curve kinetics critical for detecting partial inhibitors or covalent warheads targeting BRSK2. Radiometric assays add throughput and safety burdens.
